Sulpho-NHS (N-Hydroxysulphosuccinimide sodium salt)

Beskrivelse: Sulpho-NHS reacts primarily with primary amines to convert carboxyl groups to amine reactive sulfo-NHS esters. It is ideal for cross-linking, chemical labelling and solid support immobilisation. It is used to generate amine reactive esters from carboxylate groups and is primarily used to increase the efficiency of the carbodiimide EDC cross-linking. EDC reactions do not require Sulpho-NHS; however, its presence greatly enhances cross-linking efficiency and allows for a two-step process. EDC reacts with a carboxylate group to form an active ester, which is subject to rapid hydrolysis in aqueous buffers. The Sulfo-NHS increases the stability of the active intermediate and the resulting coupled product is identical to EDC reactions without sulpho-NHS.

T4 UvsX DNA Recombinase

Beskrivelse: T4 UvsX DNA recombinase mediates DNA strand exchange between homologous chromosomes. The protein forms a right-handed nucleoprotein complex on single ssDNA called the presynaptic filament that can search for homology in duplex DNA and pair the recombining DNA molecules to form a DNA joint molecule. This process is aided by ssDNA binding proteins and recombination mediators. The filament is then elongated by DNA polymerase with the newly synthesised strand displacing the old strand. The newly synthesised duplex DNA can then act as a template for the next cycle to achieve exponential amplification of dsDNA. <i>E.coli</i> cells expressing recombinant T4 UvsX gene.
