Beskrivelse: PLEKHM3 Antibody: PLEKHM3, also known as DAPR, is a member of the M family of Pleckstrin homology domain-containing proteins. PLEKHM3 was initially identified through chromatin immunoprecipitation and CpG microarray analysis examining proteins regulated by myocyte-enhancing factor 2. In C2C12 myoblast cells, PLEKHM3 binds to the PI3K signaling member protein kinase B in the cytosol prior to differentiation into myotubes. Following the initiation of differentiation, PLEKHM3 was also found in membrane fractions. Knockdown of PLEKHM3 expression by RNAi resulted in the inhibition of myotube formation, suggesting that PLEKHM3 is a key component required by myoblasts for orchestrating their differentiation during myogenesis.

Beskrivelse: GRTP1 Antibody: Growth hormone regulated TBC protein 1 (GRTP1) was identified through microarray analysis as a gene whose expression is regulated by growth hormone (GH). It contains the TBC signature motif of GTPase activator proteins of Rab-like small GTPases. GRTP1 mRNA is expressed at highest levels in testes, and is also present in kidney, lung, liver, and intestine. Administration of GH to mice resulted in an increase in GRTP1 mRNA in testes, but a decrease of GRTP1 mRNA in kidney and liver. GRTP1 was identified as an HIV dependency factor (HDF), suggesting that GRTP1 may be an important drug target in HIV treatment. At least three isoforms of GRTP1 are known to exist; this antibody should recognize all isoforms.

Beskrivelse: ZFP281 Antibody: ZFP281, also known as ZBP-99, is a member of a conserved family of transcription factors that interact with GC-rich promoters. It contains four Kruppel-like zinc fingers and is highly homologous to ZBP-89, another regulator of ornithine decarboxylase gene expression. ZFP281 also interacts with many other other transcription factors, such as Sp1 to repress vimentin gene transcription. ZFP281 has also been shown to control embryonic stem cell pluripotency by direct regulating specific target genes. ZFP281 phyically interacts with the transcription factos POU5F1, Sox2, and NANOG. Gene expression microarray anlysis indicated that some ZFP281 target genes were activated, while others were repressed upon knockdown of ZFP281 expression, showing that ZFP281 plays bifuctinal roles in regulating gene expression in stem cells.

Beskrivelse: The E-Z 96® Total RNA Kit is designed for isolation of total cellular RNA from up to 5×10⁶ cultured cells or soft tissues. This kit can process single or multiple samples in less than 60 minutes. Utilising HiBind® silica plate technology, the need for phenol/chloroform extractions, CsCl gradient ultracentrifugation, and precipitation with isopropanol or LiCl are eliminated. Samples are lysed in a denaturing lysis buffer which inactivates RNases. Binding conditions are adjusted and the lysate is transferred to a 96-well HiBind® RNA Plate where the RNA is purified via three wash steps. High quality RNA is eluted in RNase-free water. RNA purified using the E-Z 96® Total RNA method is ready for applications such as RT-PCR, qPCR, differential display, microarrays, and other downstream applications.
